摘要
目的:探讨水通道蛋白5(AQP5)在香烟致肺部炎症中的作用。方法:香烟提取物(CSE)刺激小鼠肺上皮细胞株MLE-12后,实时定量PCR检测其AQP5 mRNA和MUC5AC mRNA的表达。同时,用香烟烟熏BALB/C小鼠4周,检测其肺组织AQP5 mRNA和MUC5AC mRNA的表达。结果:与正常不含CSE的DMEM培养液相比较,CSE(10%)刺激MLE-12后,细胞AQP5 mRNA的表达水平降低,MUC5AC mRNA的表达水平升高。同样,香烟烟熏BALB/C小鼠1个月,其肺组织AQP5 mRNA的表达水平降低,MUC5 ACmRNA的表达水平升高。结论:CSE刺激可以降低MLE-12细胞上AQP5 mRNA的表达,香烟烟熏BALB/C小鼠1个月,其肺组织AQP5 mRNA的表达水平降低。AQP5 mRNA的下调可能通过影响其MUC5AC mRNA的表达从而参与香烟致肺部炎症的发病过程。
Objective: To investigate the role of aquaporin 5 (AQP5) in cigarette-induced pulmonary inflammation. Methods: Proper concentration of CSE was chosen to stimulate MLE-12. The expressions of AQP5 mRNA and MUC5AC mRNA were detected by real-time quantitative RT-PCR after stimulation. Meanwhile, BALB/C mice were exposed to cigarette smoke for 4 weeks, then total RNA was extracted for detection of AQP5 mRNA and MUC5AC mRNA by real-time quantitative RT-PCR. Results: After stimulation with CSE (10%), the expression of AQP5 mRNA was decreased while MUC5AC expression was increased. Meanwhile, the AQP5 mRNA expression was decreased and MUC5AC mRNA was increased in the lung of cigarette smoke exposed mice. Conclusions: CSE stimulation may lower the expression of AQP5 mRNA. The expression of AQP5 mRNA was lowered in the lung of cigarette smoke exposed mice. Down regulation of AQP5 mRNA may play a role in cigarette-smoke induced pulmonary inflammation through influencing the expression of MUC5AC.
