hNIS基因在不同甲状腺组织中表达的实验研究

Experimental study on the expression of hNIS gene in different thyroid tissues

摘要 目的观察hNIS在不同病理类型的甲状腺组织中的表达情况,进而探讨hNIS表达水平与甲状腺疾病发生、发展的关系。方法收集临床甲状腺术后患者的组织块,包括桥本甲状腺炎3例,甲状腺瘤4例、结节性甲状腺肿4例、Graves病3例、亚急性甲状腺炎2例,分别采应用RT-PCR和Western bolt的方法检测不同病理类型的甲状腺组织中hNISmRNA及蛋白质的表达水平。结果桥本甲状腺炎hNIS表达明显降低,Graves hNIS表达明显增加,亚急性甲状腺炎、结节性甲状腺肿中的hNIS表达低于正常组织,腺瘤中的hNIS表达情况高于正常甲状腺组织。结论不同病理甲状腺组织hNIS在甲状腺疾病中发挥重要的作用,hNIS表达的改变,对甲状腺疾病的发生、发展有着十分重要的意义。 Objective To observe the expression of hNIS gene in thyroid tissues of different pathologic types and to explore its relationships with thyroid disease occurrence and development.Methods Collection of clinical thyroid tissue in patients after surgery,including 3 cases of Hashimoto's thyroiditis,4 cases of thyroid tumor,4 cases of nodular goiter,3 cases of Graves,3 cases of subacute thyroiditis.The expression of hNIS mRNA and protein in different thyroid tissues of pathologic types were determined by RT-PCR and Western bolt.Results The expression of the hNIS was significantly reduced in Hashimoto's thyroiditis and was significantly increased in Graves,and it's lower than normal thyroid tissue in subacute thyroiditis and nodular goiter and it's higher than normal thyroid in thyroid tumor.Conclusions There is a big difference in the level of hNIS expression in thyroid tissues of different pathologic types.hNIS gene in thyroid diseases plays an important role.The change of hNIS expression is significant for the occurrence and development of thyroid disease.
