海尔福对铅染毒小鼠血脑生化指标干预的实验研究

摘要 目的探讨复方中药制剂(海尔福口服液)对铅中毒小鼠大脑、血液组织生化指标的干预作用。方法用浓度为2g/L醋酸铅水溶液给予小白鼠腹腔注射,每天一次,连续14d,建立铅中毒动物模型。造模后第1天,小鼠分组:大剂量治疗组(高剂量组)、小剂量治疗组(低剂量组)、模型组(即中毒不治疗组),同时设正常组(即不染毒组),然后用两个大小不同的剂量海尔福口服液给予灌胃治疗。测定实验前(铅染毒前)、实验中(中毒后,治疗前)、实验后(治疗后)各组血红蛋白(Hb),实验结束,从眼球取血,分离血清,测定各项生化指标,处死动物,取大脑,在冷冻下用生理盐水制备成10%的脑匀浆,离心取上层液放冰箱保存,待测定各项生化指标。结果高剂量组、低剂量组、模型组、正常组,脑匀浆铅依次分别为(170.96±22.29)、(154.64±13.49)、(253.49±50.90)、(28.79±7.16)×10~9mmol/L,血清铅依次分别为(105.02±53.70)、(99.20±29.86)、(270.04±61.72)、(33.20±11.52)×10~9mmol/L;治疗组明显低于模型组(P<0.01或P<0.05),正常组最低;血清ALT活性依次分别为(110.0±32.21)、(144.17±52.05)、(145.75±23.13)、(74.50±36.67)Karu,正常组和高治组明显低于模型组(P<0.05和<0.01);中毒组染毒后血红蛋白明显低于染毒前(P<0.01),治疗后治疗两组均有明显回升,而模型组没有明显变化;脑SOD活力各组依次分别为(90.54±39.26)、(81.35±21.33)、(56.65±4.71)、(133.82±68.16)u/mg.prot,模型组最低,正常组最高,两者有显著性差异(P<0.05);其他指标如脑GSH、TchE、MDA、血清尿素,尚未见明显变化。结论中药制剂对铅中毒小鼠显示出明显的疗效,降低血铅脑铅,提高血红蛋白,保护肝脏功能,提高机体抗氧化功能。 Objective To study the effects of Haierful Oral Liquid on biochemical indices of blood and brain in mice with lead poisoned.Methods To make model of lead poisoned animal with CH2COOPb liquid by ip injection,then giving the two treated groups with two different doses of Haierful Oral Liquid by pouring into stomach.At the same time,the control group(no poisoned) and the model group(poisoned but no treated) were built as control group.The two different doses of treated groups were high dose treated group and low dose treated group.The Hb of all mice were determined before,during and after experiment.When the experiment was completed,blood was got from the eyeballs and isolated and determined eversbiochemical indices of control group,model group,high treated group and low treated group.Then the mice were executed and got brains and homogenized them(10%) under the freezing surrounding,then stored in the fridge for further examination of biochemical indices.Results The lead in serum is(33.20±11.52),(270.04±61.72),(105.02±53.70),(99.20±29.86)×10~9mmol/L;The lead in brain of them is(28.79±7.16),(253.49±50.90),(170.96±22.29),(154.64±13.49)×10~9mmol/L,the indices of blood and brain are the most lowest in the control group.The treated group is evident lower than the model group(P〈0.01orP〈0.05);the vitality of SOD in brain is(133.82±68.16),(56.65±4.71),(90.54±39.26),(81.35±21.33)u/mg.prot,the control group is the highest and the model group is the lowest.There is signifi cant difference(P〈0.05);the vitality of ALT in serum is(74.50±36.67),(145.75±23.13),(110.0±32.21),(144.17±52.05)Karu;the model group is clear higher than the control group.Model group is higher than that incontrol gorup and treated group(P〈0.01and P〈0.05);comparing to the no lead poisoned.Hb of lead poisoned group is obvious lower after lead poisoned(P〈0.01).After treating,the two treated bure were all evidently uncnease,no change in model group,no signifi cant change,other indices such as GSH,TchE,MDA,serum-urea content,SOD.Conclusion Haierful Oral Liquid has signifi cant effects on the accelerating lead excretion,improving the Hb of blood,protecting the function of the liver,improving the vitality of SOD in the brain and its antioxidant function.
