可降解多孔β-TCP/rhBMP-2人工骨的骨诱导活性研究 被引量:14

Study on Osteoinductive Potential of Degradable and Porous β-TCP/rhBMP-2 Composite as a Bone Substitute

摘要 目的:检测可降解多孔β-TCP/rhBMP-2人工骨的诱导成骨能力。方法:自制可降解多孔β-TCP与rhBMP-2复合后植入45只小鼠股后肌袋作为实验组,单纯β-TCP植入对侧作对照。术后24、72小时、1、2、4、8周分别取材做组织学检查;2、4周取材的部分标本做扫描电镜观察;1、2、4周取材的部分标本做碱性磷酸酶(ALP)活性测定。结果:实验组有大量新生软骨和骨形成,随着时间的推移,生成量逐渐增多。对照组无软骨和骨生成。此外,实验组ALP活性高于对照组(P<0.01),并且随时间推移而升高(P<0.05)。结论:可降解多孔β-TCP/rhBMP-2具有良好的骨诱导作用。 Objective: To examine the osteoinductive potential of degradable and porous β-TCP/rhBMP-2 composite material. Methods: The β-TCP/rhBMP- 2 composite was obtained by combining degradable and porous β-Tricalcium phosphate (β-TCP) that was developed by the authors with recombinant human morphogenetic protein-2 (rhBMP-2) and was implanted into the muscle pouches in the thigh of mice. β-TCP alone was implanted on the opposite site as controls. After the surgery histologic studies (24 and 72 hours,1,2,4 and 8 weeks), scanning electron microscopy(2 and 4 weeks)and alkaline phosphatase assay (1, 2 and 4 weeks) were performed after implantation. Results: There was a large amount of new cartilage and bone formation within the composite material increasing with time. However,there was no new bone formation where β-TCP alone was implanted. The levels of alkaline phosphatase in the β-TCP/rhBMP-2 implants also were increasing with time and were higher than those in controls( P <0.01). Conclusion: β-TCP/rhBMP-2 possesses osteoinductive potential.
