支气管哮喘患儿外周血树突状细胞上前列腺素D_2受体改变研究 被引量:1

Change of prostaglandin D_2 receptor on monocyte-derived dendritic cells in peripheral blood in bronchial asthma

摘要 目的研究不同病期支气管哮喘(简称哮喘)患儿单核细胞衍生的树突状细胞(dendritic cells,DC)上前列腺素D2受体(DP/CRTH2)结合容量改变,探讨其对Th2极化的影响。方法随机病例对照研究。分哮喘急性发作期组32例,平均年龄(6.1±3.5)岁,缓解期组30例,平均年龄(5.8±2.6)岁,正常对照组34例,平均年龄(7.2±3.8)岁。用放射配体结合分析法测定外周血DC上DP1/CRTH2结合容量;用酶联免疫吸附试验(ELISA)法测定外周血白介素4(IL-4)、IL-5和γ干扰素(IFN-γ)。结果哮喘发作期和缓解期组DC上总结合和CRTH2受体结合容量比正常对照组显著增加,DP总结合为:(532.42±123.87)fmol/106细胞、(491.78±110.64)fmol/106细胞、(274.61±87.16)fmol/106细胞;CRTH2为:(355.66±99.19)fmol/106细胞、(316.83±81.42)fmol/106细胞、(123.74±49.27)fmol/106细胞;而DP1差异无统计学意义(P<0.01)。发作期和缓解期组Th2细胞因子IL-4、IL-5比正常对照组显著增加,IL-4为:(12.76±3.54)ng/L、(11.44±2.65)ng/L、(7.49±1.45)ng/L;IL-5为:(36.28±10.21)ng/L、(32.11±11.56)ng/L、(18.48±5.65)ng/L(P<0.01);发作期和缓解期组Th1细胞因子IFN-γ比正常对照组显著减少,IFN-γ为:(7.47±2.49)ng/L、(7.56±1.41)ng/L、(13.13±1.44)ng/L(P<0.01)。结论哮喘患儿发作期和缓解期外周血DC上CRTH2受体表达和Th2细胞因子IL-4、IL-5分泌增加。 Objective To investigate the change of prostaglandin D2 (DP) receptor on dendritic cells (DC) in exacerbation and remission phase patients with bronchial asthma(asthma) and the ability of receptor to control Th2 polarization. Methods Randomized case control clinical trial was designed. Thirty patients were randomly divided into two groups:exacerbation phase group 32 cases (6.1±3.5) years, and remission phase group 30 cases (5.8 ±2.6) years, normal control 34 cases (7.2 ± 3.8) years. Radiological binding assay was used to study the DP1/CRTH2 receptors in monocyte-derived DC in peripheral blood. The plasma levels of IL(interleukin) 4,IL-5,and INF-γ were detected by ELISA method. Results The total combining capacity and CRTH2 on DC in peri-pheral blood in exacerbation phase and remission phase group were significantly higher than those in normal controls, total combining capacity: (532.42±123.87) fmol/106 cells,(491.78±110.64) fmol/106 cells,(274.61±87.16) fmol/106 cells. CRTH2: (355.66±99.19) fmol/10^6 cells,(316.83±81.42) fmol/10^6 cells,(123.74±49.27) fmol/10^6 cells (P〈0.01). In three groups,there were no significant differences in DP1. In exacerbation phase and remission phase group, the serum levels of Th2 eytokines IL-4, IL-5 were significantly more increased than in control group, IL-4: (12. 76 ± 3.54) ng/L, (11.44±2.65) ng/L,(7.49±1.45) ng/L. IL 5: (36.28±10.21) ng/L,(32.11±11.56) ng/L, (18. 48 ±5.65) ng/L( P 〈0.01). Thl cytokine IFN 7 was significantly decreased in the above three groups,(7.47±2.49) ng/L;(71 56 ±1.41 ) ng/L, (13.13 ±1.44) ng/L( P 〈0.01 ). Conclusions There are significantly higher total combining capacity DP and CRTH2 on monocyte-derived DC in exacerbation and remission phase patients with asthma,and the serum levels of Th2 cytokine IL-4,IL-5 are significantly increased.
