老瓜头总生物碱对角叉菜胶诱导的大鼠胸膜炎的抑制作用 被引量:3

Anti-inflammatory Effects of Alkaloids from Cynanchum Komarovii Al. Iljinski on Carrageenin-induced Pleurisy in Rats

摘要 目的研究老瓜头(Cynanchum komarovii Al. Iljinski)总生物碱(ACKs)对角叉菜胶诱导的大鼠胸膜炎的抑制作用及其作用机制。方法采用吲哚美辛(10 mg.kg-1)或不同剂量的ACKs(12,6,3 mg.kg-1)于角叉菜胶(0.2mL,10 mg.mL-1)制备大鼠胸膜炎模型前30min腹腔注射给药,在造模后4h处死大鼠,测定胸腔渗出液体积,渗出液中白细胞计数,测定肺组织匀浆中丙二醛(MDA)的含量以及超氧化物歧化酶(SOD)的活性,并做肺组织形态学观察。结果吲哚美辛和ACKs各剂量均能降低胸腔液体渗出和白细胞浸润,降低肺组织中MDA的含量,升高肺组织中SOD的活性。结论ACKs对角叉菜胶制备的大鼠胸膜炎有抑制作用并减少白细胞浸润和炎性渗出,其作用机制可能与抑制活性氧对机体的损伤作用有关。 Objective To explore the anti-inflammatory effects of alkaloids from Cynanchum komarovii Al.Iljinski(ACKs) on rat carrageenin-induced pleurisy and to demonstrate the anti-inflammatory mechanism.Methods Rats were pretreated with an intraperitoneal injection of physical saline,indomethacin(10 mg·kg-1) or ACKs(12,6,3 mg·kg-1).30min later,rats were anaesthetized and irritated with carrageenin(0.2mL,10 mg·kg-1) by pleural injection.And 4h later,rats were sacrificed by exsanguination and opened the pleural cavity carefully,then moved the exudate out and measured its volume and counted the number of leukocytes in it with light microscopy.At last,the levels of MDA and SOD in lung homogenate were measured.Results Both indomethacin and ACKs could reduce the volume of exudate as well as the number of leukocytes in it and so did the levels of MDA and SOD in lung homogenate.Conclusion The anti-inflammatory effects of ACKs on rat carrageenin-induced pleurisy may lie in the infiltration of leukocytes and scavenging of radical free oxygen and oxide.
