摘要
目的观察合欢皮总皂苷低剂量组(5.25mg·kg-1)和高剂量组(26.25mg·kg-1)对大鼠血液循环系统及血液常规的影响。方法SD雄性大鼠,实验分为低剂量组(5.25mg·kg-1)、高剂量组(26.25mg·kg-1)及生理盐水(NS)对照组,生物信号记录系统记录给药后1h的血压、心电图(ECG)、脉搏及血常规的变化。动脉插管测定血压针式电极测定Ⅱ导联心电图,尾动脉记录脉搏波,细胞计数法测定血液常规。结果(1)血压:与正常对照组相比,低剂量组无显著变化,高剂量组明显升高。(2)脉搏:与正常对照组相比,低剂量组搏动频率、幅值和上升斜率均无显著差异;高剂量均增加。(3)心电图:与正常对照组相比,低剂量组无显著变化,高剂量组P-R间期和Q-T间期延长,P波、QR波S和T波幅值升高。(4)血液常规:低剂量对血常规无显著影响,高剂量使白细胞计数、血小板计数和血红蛋白含量明显下降。结论高剂量合欢皮总皂苷对大鼠血液及循环系统有一定的毒性作用。
OBJECTIVE To observe the effect caused by the low dose(5.25mg·kg-1)and the high dose(26.25mg·kg-1)of the Silktree Albizia on the blood and the circulation system of rats.METHODS The rats were divide into three groups,including low dose group(5.25mg·kg-1),high dose group(26.25mg·kg-1)and the cotrol group(NS)respectively.after administration 1h,the change of blood presure(BP)by arterial cannulation,electrocardiacgram(ECG)by needle node,pulse wave by caudal artery were recorded by the biologic signal system,and calculate the blood cells and the hemoglobin by counting method.RESULTS(1)Blood pressure:Compared with the control group,no difference was found in the low-dose group,and the blood pressure in the high dose group showed a significent increased.(2)Pulse:Compared with the control group,wave frequency,width value and ascensus slope rate in low dose group have no difference,while these index in high dose group were increased.(3)ECG:Compared with the control group,low-dose group found no obvious change in the index,while in the high dose group,the duration of P-R,QRS and Q-T was prolonged,and the amplitude of P wave and T wave showed increased.(4)Bood index:Compared with the control group,no significent difference was found in the low dose group,but in the high dose group,the number of the white blood cells(WBC),red blood cells(RBC),and the hemoglobin content were decreased.CONCLUSION The high dose of total saponin of the Silktree Albizia have certain toxicity to the rat's blood and the circulation system.
