摘要
目的探讨经顶部入路预置管二次注射自体动脉血建立大鼠尾状核脑出血模型的可行性。方法12对SD雄性大鼠,随机分为预置管组(12只)和对照组(12只)。预置管组大鼠在立体定向仪辅助下经顶部入路穿刺尾状核中心并置管,24h后取股动脉血50ul,分2次缓慢注入尾状核(10ul/min);对照组大鼠在立体定向仪辅助下置管后立即分2次注射自体动脉血50ul。测量两组大鼠血肿形态和体积并进行统计学分析。结果两组大鼠左侧尾状核均可见血肿形成,预置管组血肿形态较规则,呈圆形,占位效应较明显。预置管组平均血肿体积为(25.1±0.5)ul,显著大于对照组的(24.2±0.7)ul(P〈0.05),血肿量离散系数显著低于对照组(2.0%对2.9%,P〈0.05)。结论预置管二次注射自体动脉血可建立较为稳定的大鼠脑出血动物模型。
Objective To investigate the feasibility of establishing a intracerebral hemorrhage model in rats by injecting autologous arterial blood into caudate nucleus twice through parietal approach via preset canula. Methods Twelve pairs of male SD rats were randomly allocated into either a preset canula group (n = 12) or a control group (n = 12). A canula was inserted through parietal approach by puncturing caudate nucleus center and indv, elling a canula under the aid of a stereotactic apparatus in the preset canula group. After 24 hours, 50 uL of autologous arterial blood from femoral artery was collected, and was injected slowly into cap,date nucleus (10 uL/min) twice; immediately after a canula was inserted under the aid of a stereotactic apparatus in the control group, 50 uL of autologous arterial blood was injected twice, The shape and volume of hematoma in both groups were measured and a statistical analysis was conducted. Results Hematoma formation could he observed in the left caudate nucleus of rats in both groups. The hematoma shape was more regular, round in shape, and the mass effect was more obvious in the preset canula group. The mean hematoma volume in the preset canula group was 25.1 - 0.5 uL, and it was significantly greater than 24.2 ± 0. 7 uL in the control group (P 〈0.05). The dispersion coefficient of hematoma volume was significantly lower than the control group (2.0% vs. 2.9%, P 〈 0.05). Conclusions The injection of autologous arterial blood twice via preset canula may establish a more stable intracerebral hemorrhage model in rats.
