摘要
在红掌组织培养过程中,外植体易发生褐变,严重阻碍了红掌组织培养的正常进行。现以红掌‘红粉佳人’(Anthuriumandraeanum cv.Sonate)品种为试材,取健康植株幼嫩叶片为外植体,对其愈伤组织培养过程中褐变现象进行研究。研究不同抗氧化剂、吸附剂(柠檬酸、抗坏血酸VC、叶酸、聚乙烯吡咯烷酮PVP、活性碳AC)以及不同培养基硬度的防褐化效果。结果表明:使用硬度为5 g/L琼脂的MS培养基,并添加6-BA1.0 mg/L+2,4-D0.2 mg/L+聚乙烯吡咯烷酮(PVP)0.2%可有效控制褐变并获得较高的愈伤诱导率。
Tissue oxidative browning is a big problem which interferes with normal propagation of Anthuriurn andraeanum tissue culture. The young leaves of Anthurium andraeamum cv. Sonate were used as explants in order to study the browning of tissue culture. The treatments included two adsorbents: active carbon, PVP, three antioxidants: citric acid, VC, folic acid, and five culture medium. Results showed that the best treatment for the browning control of explanted tissue is the explants were cultured in MS with agar(5 g/L), added with 6-BA(1.0 mg/L) ,2,4-D(0. 2 mg/L),and PVP (0.2%).
