摘要
目的研究聚ADP核糖聚合酶-1(PARP-1)和Caspase-3在乳腺癌中的表达及其与临床特征的关系。方法采用免疫组化技术,检测277例乳腺癌和11例乳腺正常组织石蜡包埋标本中PARP-1及Caspase-3的表达,分析正常乳腺与乳腺癌组织中两者表达与临床参数的相关性,以及两者间的相关性。结果在277例乳腺癌组织中,PARP-1阳性表达率(62.09%,182/277)显著高于正常乳腺组织(27.27%,3/11)(P=0.009)。乳腺癌中有淋巴结转移者PARP-1阳性表达率(76.47%,130/170)高于无淋巴结转移者(48.59%,52/107),表明PARP-1与淋巴结转移相关(r=-0.286,P<0.001)。同时PARP-1在乳腺癌中的表达与组织学分级相关(r=0.018,P=0.002)。Caspase-3在正常乳腺组织的阳性表达率(72.73%,8/11)高于乳腺癌(42.24%,117/277)(P=0.046)。Caspase-3与PARP-1的表达在乳腺癌组织中呈负相关(r=-0.722,P<0.001)。结论PARP-1表达上调可能与乳腺癌的恶性生物学行为有关,Caspase-3表达下调可能与乳腺癌的发生、发展有关。
Objective To investigate the expressions of PARP - t and Caspase - 3 in normal breast tissues and breast cancer, and their relationships with the clinical features. Methods PARP - 1 and Caspase -3 expressions were determined immunohistochemieally on paraffin - embedded specimens of 277 breast cancer and 11 breast normal tissues. Relationships between expressions of PARP - 1 and Caspase - 3 and among the clinical features of breast normal tissues and breast cancer, as well as the relevance of PARP - 1 and Caspase - 3 in these tissues were analyzed. Results The positive rate of the expression of PARP - 1 in breast cancer ( 62.09%, 182/277 ) was significantly higher than that in normal breast ( 27.27% , 3/11 ) ( P = 0.009 ). The positive rate of the expression of PARP - 1 in the lymph node matastasis group ( 76.47%, 130/170) is higher than that in the no lymph node matastasis group (48.59%, 52/107 ). PARP - 1 expression was significantly correlated with the lymph node metastasis ( ( r = - 0. 286, P 〈 0. 001 ) and the histological grade ( r = 00180, P = 0.002 ). The positive rate of the expression of Caspase -3 in normal breast (72.73%, 8/11 )was significantly higher than that in breast cancer ( 42.24%, 117/277 ) ( P = 0.046 ). PARP - 1 had negative correlation with Caspase - 3 in breast cancer tissues ( r = - 0. 722, P 〈 0.001 ). Conclusion Increased expression of PARP - 1 may be related to the malignant biological behavior of breast cancer. Decreased expression of Caspase -3 may be involved in the occurrence and development of breast cancer.
