川芎嗪对脓毒症致兔急性肾损伤的保护作用 被引量:4

摘要 目的研究脓毒症致急性肾损伤(AKI)的发病机制,探讨川芎嗪(TMP)对脓毒症致AKI实验动物的肾脏保护作用。方法36只雄性新西兰大白兔随机分成3组:空白对照组(C组)、脓毒症模型组(M组)和TMP干预组(P组)。静脉注射脂多糖(LPS,1mg/kg)制备脓毒症兔实验动物模型,P组在静脉注射LPS后相继静注TMP(60mg/kg),实验过程中测定实验动物尿量,实验结束时测定血半胱氨酸蛋白酶抑制剂C(Cys-C)浓度;处死实验动物后提取肾脏组织,测定肾脏含水率,取肾组织进行病理学观察,用免疫组化方法检测肾组织vWF(vWF)。结果与M组比较,C组和P组肾脏组织含水率较低(P<0.05)、4h尿量明显增多(P<0.01)、血Cys-C浓度也较低(P<0.05),免疫组化法测定的vWF、HE染色和电镜观察,P组实验动物肾脏病理改变程度较M组轻。结论脓毒症可导致AKI,在早期以肾小球滤过率降低为主,TMP对脓毒症实验动物AKI具有保护作用。 Objective To study the mechanism of the sepsis-induced acute kidney injury(AKI) and to explore the protective effects of tetramethylpyrazine (TMP) on the AKI.Methods Thirty-six male New Zealand white rabbits were randomly divided into three groups:normal control group (C group),sepsis model group (M group) and TMP group (P group).Group M and P underwent the intravenous injection of lipopolysaccharide (LPS,1 mg/kg),and group P was treated with TMP (60 mg/kg),immediatelty after LPS was given.Urine output and blood cystatin C (Cys-C) concentration were measured.Kidney tissues were extracted after execution and pathological changes were observed after renal water content was detected,and vWF were determined by immunohistochemistry method.Results Compared with group M,the other two groups (C and P) were reported of lower water content in kidney tissue (P〈0.05) and serum Cys-C concentration (P〈0.05),and the fourth-hour urine volume increased with statistical significance (P〈0.01).Pathological changes observed through HE staining,electron microscopy and the results of immunohistochemistry showed that kidney damage in group P was lighter than group C.Conclusion Sepsis can lead to the AKI with damaged glomerular filtration rate in the early stage and TMP may exert protective effects on reducing glomerular damage in New Zealand white rabbits.
