摘要
目的建立高效液相色谱-蒸发光散射检测法,对宁夏栽培与野生柴胡中柴胡皂苷a、d的含量进行比较研究。方法 Agilent ZORBAX XDB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-水(43∶57)为流动相,流速1.0 ml/min,柱温:30℃,Agilent ELSD 2000ES检测器,漂移管温度:97℃,气速:2.6 L/min。结果柴胡皂苷a、d的线性范围分别是0.2002-8.008μg(r=0.999 5),0.261-10.45μg(r=0.998 5);平均回收率分别为97.4%、97.5%;RSD分别为1.5%、2.5%。宁夏栽培与野生柴胡中柴胡皂苷a、d的含量之和范围在0.92%-1.61%。结论宁夏栽培柴胡中柴胡皂苷a、d的含量之和与野生柴胡基本一致。本法简便、准确、重复性好,可作为柴胡药材质量检测的方法。
Objective To establish a method of HPLC-ELSD for determination of saikosaponins a,d in wild and cultured Bupleurum in Ningxia.Methods An Agilent ZORBAX XDB-C18(250 mm×4.6 mm,5 μm) column was used at 30 ℃,acetonitril-water(43:57) was used as the mobile phase.The flow rate was 1.0 ml/min.Agilent ELSD 2000ES detector as the detector.The drift tube temperature was 97 ℃,the gas flow rate was 2.6 L/min.Rusults The linear ranges of saikosaponin a,d were 0.2002-8.008 μg(r=0.9995),0.261-10.45 μg(r=0.9985).The average recoveries were 97.4%,97.5% for saikosaponins a,d,respectively.The ranges of the content of saikosaponins a and d in wild and cultured Bupleurum in Ningxia were 0.92%-1.61%.Conclusion The total content of saikosaponins a and d in cultured Bupleurum in Ningxia are basically identical with wild Bupleurum.This method is simple,sensitive and reliable.It can be used for the quality control of Bupleurum.
