大肠癌细胞PARG、PARP与AKT磷酸化状态的关系 被引量:2

摘要 目的:聚(腺苷二磷酸核糖)水解酶[Poly(ADP-ribose)glycohydrolase,PARG]可通过影响聚(腺苷二磷酸核糖)聚合酶[Poly(ADP-ribose)polymerase,PARP]和蛋白激酶B[protein kinase B,AKT]磷酸化在炎症中发挥重要作用,但在肿瘤中的作用却不清楚。本研究则对大肠癌细胞系LOVO中PARG、PARP、NF-κB和AKT磷酸化的相互关系进行了初步探讨。方法:Western blot法检测LOVO细胞PARG、PARP、NF-κB、AKT和PI-AKT473的蛋白表达。以丹宁酸为PARG抑制剂。结果:PARG抑制剂丹宁酸处理组LOVO细胞PARG、PARP、NF-κB表达降低,而PI-AKT473表达则增强,较对照组(无丹宁酸处理组)比较差异均具有显著意义(P=0.0068,P<0.0001,P<0.05,P=0.0008)。且PARG和PARP蛋白的表达呈正相关(r=0.8238,P<0.05),PARG和PI-AKT473的蛋白表达呈负相关(r=-0.8190,P<0.05)。结论:在大肠癌细胞株中,抑制PARG可以下调PARP和NF-κB,并可增强AKT的磷酸化。其在肿瘤浸润转移中,可能具有重要作用。 Objective:Poly(ADP-ribose) glycohydrolase(PARG) plays an important role in the inflammation via regulating Poly(ADP-ribose) polymerase(PARP) and AKT phosphorylation,but it is not clear that how it in tumour.The current study was desiged to study the relationship between PARG,PARP,NF-κB and AKT phosphorylation in colorectal carcinoma cell lines LOVO.Methods:Western blot was used to test the expression of PARG,PARP,NF-κB,AKT and PI-AKT473.Gallotannin(GLTN) was served as PARG inhibitor.Results:The expression of PARG,PARP and NF-κB in gallotannin-treated LOVO was weaker than that in the control groups(gallotannin-untreated groups)(P=0.0068,P0.0001,P0.05).While the expression of PI-AKT473 was increased in gallotannin-treated groups(P=0.0008).A positive correlation was found between PARG expression and PARP expression(r=0.8238,P0.05).But the expression of PARG was independently and inversely correlated with PI-AKT473 expression(r=-0.8190,P0.05).Conclusion:The data suggest that PARG inhibition may downregulate PARP,NF-κB and enhance AKT phosphorylation in colorectal carcinoma cells.PARG probably plays a role in the infiltration and metastasis in tumour.
