摘要 以薯蓣皂苷元的得率为指标,采用高效液相色谱法测定其含量,通过单因素试验考察了影响黄姜总皂苷提取和水解的主要因素。结果表明,样品中薯蓣皂苷元适宜的色谱分析条件为色谱柱Hyper-sil ODS2(250 mm×4.6 mm,5μm),甲醇为流动相,体积流量1.0 mL/min,进样量20μL,柱温常温,检测波长208 nm,此时薯蓣皂苷元在0.05~2.0 mg/mL峰面积与其质量浓度具有很好的线性关系(r=0.999 4);黄姜总皂苷提取的最佳工艺条件为乙醇体积分数70%,料液比1∶8,回流提取3次,每次2 h;黄姜总皂苷水解的最佳工艺条件为硫酸浓度2 mol/L,硫酸用量为30 mL,水解时间4 h。本方法简便易行,适用于工业化生产。 The extraction and hydrolysis conditions of total saponins in D.zingiberensis were established by single factor test with the yield of diosgenin as index.The contents of diosgenin were determined by HPLC.The chromatographic column was a Hypersil ODS2(250 mm×4.6 mm,5 μm).Methanol was used as mobile phase.The flow rate was set at 1.0 mL/min.The injection volume was 20 μL for each analysis.The column temperature was ambient and the detection wavelength was set at 208 nm.Results showed that the linear range of calibration curve was 0.05-2.0 mg/mL(r=0.999 4).The optimum extraction conditions were developed as follows: ethanol concentration 70%,ethanol amount 8 times,backflow extraction 3 times,and 2 h for each time.The optimum hydrolysis conditions were also developed as follows: sulphuric acid concentration 2 mol/L,sulphuric acid amount 30 mL,hydrolysis time 4 h.The method was simple and applicable to industrial production.
