用荧光原位杂交(FISH)方法研究人类卵巢癌和胃癌细胞系17号染色体拷贝数变化 被引量:2

摘要 目的研究载有多种癌变相关基因的17号染色体在不同器官、不同组织类型肿瘤单个细胞内拷贝数的变化规律。方法以正常外周血淋巴细胞为对照,采用FISH方法检测各细胞株17号染色体拷贝数。结果17号染色体在各细胞株单个细胞中拷贝数表现为:SKOV3以六倍体为主,占48%,七倍体率占23%;3AO以三倍体为主,占93%;OV2以四倍体为主,占67%;MGC803以六倍体为主,占64%;BGC823以二倍体为主,占83%;SGC7901以三倍体为主,占80%;PAMC82以五倍体为主,占78%。结论各细胞系均无17号染色体缺失,反呈扩增趋势。 Objective To study the copy number change at chromosome 17 in different tumor cell lines.Methods Fluorescence in situ hybridization (FISH) was used to test the copy number in individual ovarian cancer and gastric cancer cells in vitro .The normal peripheral lymphocytes were used as controls. Results chromosome 17 in SKOV3 is hexaploid (48%),seven ploid(23%),tirploid (93%)in 3AO,tetropoid (67%)in OV2,hexa ploid (64%) in MGC803,biploid(83%) in BGC823.Conclusion There is no chromosome 17 deletion in most cell lines studied. On the contrary, it showed a tendency of amplification.Further study is needed for the understanding of the significance of the results.
