摘要
目的:探讨microRNA143、microRNA145与肝外胆管癌的关系以及microRNA143、microRNA145对胆管癌细胞株QBC939细胞周期及凋亡的影响。方法:提取11对肝外胆管癌组织标本和正常胆管组织标本的RNA,实时荧光定量PCR(realtimePCR)检测microRNA143、microRNA145的表达;将microRNA143、microRNA145质粒转染人胆管癌细胞株QBC939,分别以流式细胞仪和噻唑蓝(MTT)的方法检测转染前后细胞周期、凋亡及生长情况。结果:相对正常胆管组织,胆管癌组织microR-NA143、microRNA145表达率分别为0.048、0.035(P<0.05)。较对照质粒组,microRNA143组、microRNA145组G1期细胞百分比明显增多[(66.08%±1.93%、65.57%±1.85%)vs(42.87%±1.37%)]、S期细胞百分比明显减少[(19.16%±1.15%、21.40%±1.65%)vs(38.76%±1.83%)]、细胞凋亡率增加[(18.49%±2.26%、16.89%±2.47%)vs(4.68%±1.65%)]、细胞生长受抑制(P<0.05)。结论:microRNA143、microRNA145在肝外胆管癌中低表达,它们可能与胆管癌的发生有关;使胆管癌细胞株QBC939microRNA143、microRNA145表达上调,能够抑制细胞增殖,诱导细胞凋亡和周期阻滞。
Objective:To explore the effects of microRNA ld3,microRNA 145 transfection on biological behaviors of cholangiocarcinoma cell line QBC939,and the relationship between microRNA 143,microRNA 1,45 and extrahepatic cholangiocarcinoma~ Methods: RNA of 11 pairs of specimens of extrahepatic cholangiocarcinoma and normal bile duct was extracted,and then the expression of microRNA 143 and microRNA 145 was detected by real-time PCR. Expression plasmid of microRNA 143 (or microRNA 145) was transiently transfected into human cholangiocarcinoma cell line QBC939, and then cell proliferation, cell cycle and cell apoptosis were determined by MTY and flow cytometry assay. Results:Compared with normal bile duct,the relative expression ratio of microRNA143 and microRNA145 in extrahepatic cholangiocarcinoma respectively was 0.048,0.035(P 〈 0.05). Compared to control group, cholangiocarcinoma cell growth was effectively inhibited in microRNA143 group and microRNA145 group,the percentage of cells in G1 phase was increased [ (mieroRNA143 group and microRNA145 group:66.08%±1.93% and 65.57%±1.85%)vs(eontrol 42.87%±1.37%)] ,the percentage of cells in S phase was decreased[ (19.16%±1.15% and 21.40%±1.65%) vs (control 38.76%±1.83%)]and the ratio of cell apoptosis was increased[ (18.49%±2.26% and 16.89%±2.47%)vs 4.68%±1.65%] (P 〈 0.05). Conclusion:The expression of microRNA143 and microRNA 145 is down-regulated in extrahepatic cholangiocareinoma, and they may correlate with tumorigenesis. With up-regulation of microRNA 143 or microRNA 145 expression in eholangioeareinoma cell line QBC939, cell proliferation can be inhibited, cell cycle can be arrested and cell apoptosis can be increased.
