摘要
目的:了解降钙基因相关肽(calcitonin gene-related peptide,CGRP)浓度在单侧睾丸扭转复位后对侧睾丸组织中不同时段的变化,阐明CGRP与对侧睾丸早期损伤之间的联系。方法:将30只青春期前SD大鼠随机分为假手术组、扭转复位组和扭转切除(不复位)组,每组10只。假手术组作为对照;扭转复位组和扭转切除组依Turner法建立左侧睾丸扭转模型,在扭转4h后,前者行复位固定术,后者行切除术。三组分别于实验完成后4h、24h各处死大鼠5只,取右侧睾丸组织行CGRP及乳酸脱氢酶(actatedehydrojgenase,LDH)浓度检测。结果:扭转切除组4h与24h时CGRP浓度分别为(712.4±23.1)μmol/L与(733.0±30.0)μmol/L,LDH浓度分别为(417.2±25.9)U/L与(458.1±23.5)U/L;而扭转复位组的CGRP在4h与24h时分别为(852.3±33.9)μmol/L与(861.7±29.8)μmol/L,LDH分别为(690.0±28.8)U/L与(706.7±32.3)U/L;显示CGRP与LDH浓度在扭转复位组均有不同程度升高(P<0.05),但各指标在扭转切除组与假手术组的相比中未显示明显的统计学差异。结论:在单侧睾丸扭转复位后,对侧睾丸组织可能存在缺氧状态,CGRP的反应性升高可能起一定保护作用。
Objective: To understand the changes of CGRP concentration on contralateral testis of SD rats with unilateral testicular torsion in different time intervals,and explain the association between CGRP and the injury on contralateral testis.Methods: Thirty prepubertal Sprague-Dawley rats were randomly divided into 3 groups,(10 rats in each group).The rats of sham operation group underwent a sham operation of the left testis under general anaesthesia.Those of torsion/detorsion group and the torsion/orchiectomy group underwent a similar operation but the left testis was rotated 720 °clockwisely for 4 h,maintained by fixing the testes to the scrotum,then detorsion was performed in the torsion/detorsion group,while orchiectomy was performed in the torsion/orchiectomy group.The right testes of five rats from each group were removed and collected at 4 hour and 24 hour intervals respectively after surgery.Then the biochemical parameters,such as calcitonin generelated peptide (CGRP) and lactate dehydrogenase (LDH) were evaluated.Results: In torsion/orchiectomy group,the CGRP concentrations at 4 h and 24 h intervals were (712.4 ± 23.1) μmol/L and(733.0 ±30.0)μmol/L,the LDH concentrations were(417.2 ±25.9)U/L and(458.1 ± 23.5)U/L ;while in torsion/detorsion group the CGRP concentrations were(852.3 ± 33.9)μmol/L and (861.7 ±29.8)μmol/L,the LDH concentrations were(690.0 ±28.8)U/L and(706.7 ±32.3)U/L;which showed the biochemical parameters in the torsion/detorsion group increased in different degrees (P 0.05).However,there were no significant differences of each index between the sham operation group and torsion/orchiectomy group.Conclusion: These results suggest that there be a hypoxia condition on contralateral testis after unilateral testicular torsion/detorsion,and CGRP may play definte protective role on contralateral testes.
