摘要
目的:建立高效液相色谱-紫外检测(HPLC-UV)法测定自发性高血压大鼠(SHR)血浆中去甲肾上腺素(NA)的含量。方法:色谱柱为C18,流动相为甲醇-0.15mo·lL-1磷酸二氢钾缓冲液(1∶9,V/V,pH6.0),检测波长为280nm,柱温为室温,流速为1.0mL·min-1。结果:NA检测浓度线性范围为0.5~40μg·L-1(r=0.9999),最低检测浓度为0.05μg·L-1,平均回收率为97.49%,RSD小于3.81%。结论:本方法线性、精密度、回收率良好,灵敏度高,操作简便,可用于NA的药动学及相关研究。
OBJECTIVE: To establish an HPLC-UV method for the content determination of noradrenaline (NA) in plasma of spontaneous hypertension rats (SHR).METHODS: The determination was performed on C18 column with mobile phase consisted of methanol-0.15 mol·L-1 potassium dihydrogen phosphate buffer (1 ∶ 9,V/V,pH 6.0) at flow rate of 1.0 mL·min-1.The detection wavelength was set at 280 nm and column temperature was room temperature.RESULTS: The linear range of NA was 0.5~40 μg·L-1 (r=0.999 9) with an average recovery of 97.49% (RSD3.81%).The detection limit was 0.05 μg·L-1.CONCLUSION: The method is sensitive and simple with good linearity,precision and recovery for pharmacokinetic study and relevant research of NA.
