摘要
建立了高效阴离子交换色谱-积分安培检测器法测定乳果糖口服溶液中乳果糖及其5种有关物质。采用CarboPac PA20色谱柱及预柱,以250mmol/L氢氧化钠溶液-100mmol/L乙酸钠溶液-水(3︰3︰96)为流动相,金电极为工作电极,银-氯化银电极为参比电极。乳果糖及其5种有关物质分别在3.2~28.8、0.32~2.88、0.10~0.86、0.22~1.98、0.48~4.32和0.03~0.29μg/ml浓度范围内线性关系良好,检测限为0.02~0.04ng,回收率为97.5%~103.4%,RSD为0.5%~2.2%。
An high-performance anion-exchange chromatography with integrated pulsed amperometric detection was established for the determination of lactulose and its five related substances in lactulose oral solution. A CarboPac PA20 analytical column and a guard column were used with 250 mmol/L sodium hydroxide solution-100 mmol/L sodium acetate solution-water (3 : 3 : 96) as the eluent. A gold electrode was used as the working electrode with the silver-silver chloride electrode as reference electrode. The calibration curves of lactulose and its five related substances were linear in the concentration ranges of 3.2 - 28.8, 0.32 - 2.88, 0.10 - 0.86, 0.22 - 1.98, 0.48 - 4.32 and 0.03 - 0.29 ug/ml, respectively. The detection limit was 0.02 - 0.04 ng. The recoveries were 97.5 % - 103.4%, with RSDs of 0.5 % - 2.2 %.
