摘要
采用反相高效液相色谱法(RPHPLC)测定胰岛素聚乳酸微球的含量。在ODS柱上,以乙腈0.01%三氟乙酸(30∶70)为流动相,于276nm处进行检测。方法的平均回收率为99.93%,RSD=0.24%(n=3)。线性范围在0.1064~2.128mg/ml。
RP HPLC analysis of insulin in poly D,L lactic acid (PDLA) microspheres was presented in this study. Separation and determination of insulin in PDLA microspheres were achieved on a Shim pack CLC ODS column (150 mm×6 mm ID, maintained at room temperature) with detection wavelength of 276 nm. The composition of the mobile phase was acetonitrile and 0.01% trifuroacetic acid (30∶70). Good linearity was obtained from 0.1064 to 2.128 mg/ml of insulin with C =4.491×10 -4 A -2.461×10 2( r =0.9999). The average recovery was 99.93%, with RSD of 0.24% ( n =3).
