摘要
目的探讨急性压力超负荷时心脏局部碱性成纤维细胞生长因子(bFGF)和肾素血管紧张素系统(RAS)在心肌肥厚发病中的作用。方法采用Wistar大鼠腹主动脉部分结扎组(OG),并与假手术组(CG)对比检测6个时相点(0.5小时、4小时、1天、5天、10天和30天)的左心室重量指数(LVWI)、心肌局部bFGF和RAS。结果(1)OG中LVMI第5天增高(P<0.05),第10天和第30天更明显(P<0.01)。(2)OG中心肌bFGF免疫反应特异性染色强度增加,半定量显示5天升高(P<0.05),30天基本恢复正常(P>0.05);bFGF mRNA表达1天升高(P<0.05),其后类似bFGF。(3)心肌血管紧张素转换酶(ACE)活性和血管紧张素Ⅱ(AngⅡ)水平在术后0.5小时即升高(P<0.05),4小时明显增高(P<0.01),峰值在30天;心肌ACE蛋白及ACE mRNA表达均在术后4小时明显增高(P<0.05或<0.01),其后逐渐升高,峰值在30天;5天及其后,LVMI与ACE蛋白及ACE mRNA表达数量呈正相关。结论本研究提示,心肌bFGF参与急性压力超负荷致心肌肥厚的早期阶段,而心肌局部RA?
Objective To investigate the role of intracardiac renin angiotensin system (RAS) and bFGF inpathogenesis of myocardial hypertrophy. Methods Left ventricular mass index (LVMI), intracardiac RAS andbFGF were determined at 0.5 h, 4h, 1day (1d), 5 days (5d), 10 days (10d) and 30 days (30d) afterabdominal aorta constriction in rats (OG) and compared with false operation rats (CG). Results (1) LVMIgradually increased from 5d to 30d in OG (P<0.01); (2) Counts of intracardiac bFGF immune reaction andbFGF mRNA expression were increased at 5d, were essentially normalized at 30d; (3) Intracardial ACE activity,angiotensin Ⅱ (Aug Ⅱ) concentration, ACE protein and ACE mRNA expression were propessively risen from 0. 5h or 4h to 30 d; (4) LVMI was positively correlated with the level of ACE protein and ACE mRNA expressionfollowing 5d. Conclusion bFGF may only play an importannt role in early stage of cardiac hypertrophy inducedby pressure overload while intracardiac RAS may take part in various stages of the myocardial hypertrophy.
