摘要
为探讨输精管结扎对生精细胞中凋亡调节基因Bcl-2、Bax蛋白表达的影响,应用免疫组织化学法检测了大鼠输精管结扎后2~12周睾丸生精细胞BCl-2、BCX的蛋白表达情况。结果发现,输精管结扎后总的生精细胞中Bcl-2表达阳性者减少,而Bax表达阳性者增加。在结扎组,平均300个精原细胞和300个精母细胞中,Bcl-2阳性细胞数分别为45.1±14.9、71.3±15.7,而在假手术组,阳性细胞数为52.7±12.1、81.1±14.3.结扎组较假手术组均有显著减少(P<0.05)。在结扎组,平均300个精原细胞和300个精母细胞中,Bax阳性细胞数分别为84.2±19.4、49.9±17.3,而在假手术组.阳性细胞数为65.9±17.0、42.9±17.3.结扎组较假手术组均有显著减少(P<0.05)。表明输精管结扎后凋亡抑制基因BCl-2的表达有减少趋势,凋亡促进基因Bax的表达有增强趋势,以促进生精细胞的凋亡。
Immunohistochemical method was used to detect the expression of Bcl - 2 and Bax protein in the testis of vasectomized and sham - operated adult rats during and at the end 0f the l2th postoperative week. The results showed that the total number of Bcl-2 positively stained cells after vasectomy declined. In vasectomized rats .on an average in each 300 spermatogonia and each 300 spermatocytes, the number of Bcl - 2 positively stained cells was 4.1±14. 9 and 71.3±15. 7 separately,while in the sham - operated group,the number was 52. 7±12. 1 and 81. 1±14. 3 separately. The number of Bcl - 2 positive cells in vasectomized group was sigificantly decreased (P<0.05). After vasectomy, the number of Bax positive cells in each 300 spermatogonia and each 300 spermatocytes was 84. 2±19. 4 and 49. 9±17. 3 respectively .while in the sham-operated group.the number was 65.9 ±17. 0 and 42. 9 ±17. 3 respectively; the number of Bax positive cells was increased significantly (P<0.05 ). These findings demonstrate that vasectomy may decrease the expression of Bcl-2 protein nnd increase the expression of Bax protein .therefore enhance apoptosis of spermatogenic cells after vasectomy.
