摘要
以抗氧化活性为示踪,采用超声辅助提取法,在单因素筛选的基础上,对超声提取过程的影响因素进行研究,以多酚质量和清除ABTS自由基能力为双响应因子,进行3因素3水平的响应面法试验设计,并对提取工艺进行优化。结果表明,最佳提取工艺条件:乙醇体积分数28.62%,提取时间30.90 min,液料比19.96 mL.g-1,提取次数3次。在最佳提取工艺条件下,2.0 g蒙古栎叶片原料中多酚质量为84.88 mg,ABTS自由基清除能力为0.79 mol.g-1。蒙古栎叶片多酚清除DPPH自由基的EC50值为223 mg.L-1,高于BHA、BHT、VC和VE等4种合成抗氧化剂。
The influencing factors on ultrasound extraction of polyphenols from the leaves of Quercus mongolica Fischer were studied by the single factor experiment using the antioxidant activity as tracing.Response surface methodology(RSM) required for three factors at three levels was applied to optimize the extraction process for polyphenols in terms of the content of polyphenols and the ability to scavenge ABTS radical cation.The optimal conditions for the extraction of polyphenols were obtained as volume fraction of ethanol 28.62%,ultrasound extraction time 30.90 minutes,solid-to-liquid ratio 19.96 mL·g-1,and extraction for three times.Under the optimal conditions,the content of polyphenols in 2 grams of leaves was 84.88 mg and the ability to scavenge ABTS was 0.79 mol·g-1.EC50 of DPPH free radical scavenging ability of the polyphenols was 223 mg·L-1,which was higher than that of BHA,BHT,VC,and VE.
