摘要
利用AFLP技术,分析比较雌雄哲罗鱼Hucho taimen(Pallas)之间的遗传差异。采用两个酶切组合,共45对引物组合对雌雄个体进行扩增。结果表明,电泳检测条带清晰丰富,两个酶切组合分别扩增出958和971个位点,多态性比例分别在4.76%~62.26%和6.98%~37.50%,15个位点在雌雄个体中的比例存在很大差异,聚类分析结果显示雌鱼聚在一起,雄鱼聚在一起,未发现雌雄特异位点。研究积累了大量AFLP分析数据,可为哲罗鱼性别的进一步研究奠定基础。
AFLP technique was used to screen the sex-related DNA markers through analyzing DNA differences between female and male of H.taimen(Pallas).Two groups of restriction enzymes with 45 pairs of AFLP primer were selected to detect the genome DNA of the female and male individuals by AFLP analysis.The results showed that the amplified results from electrophoresis were clear and rich.Totally 958 and 971 bands were obtained by two groups,respectively,the ratio of polymorphic loci was 4.76%-62.26% and 6.98%-37.50%.15 of the loci showed apparently different distributions between female and male groups,the cluster analysis indicated that the female and male group clustered,respectively,no sex-specific loci was found.The data obtained by the study provided a base for the further research of sex determination in taimen.
