摘要
目的建立适合基因表达谱研究的小鼠结肠癌肝转移模型。方法将小鼠结肠癌细胞(C26)悬液接种于BALB/C小鼠脾内,分为切脾组和保脾组,观察小鼠生存时间及腹腔内肿瘤生长情况。结果切脾小鼠自然生存期11~15d,平均(13.05±0.71)d,保脾小鼠自然生存期9~15d,平均(11.64±1.49)d。小鼠肝脏转移率均为100%。切脾小鼠肝各叶完全被肿瘤占据,保脾小鼠脾内肿瘤巨大,肝内转移多为散在的米粒大小瘤结节转移。结论用于基因表达谱研究的小鼠结肠癌肝转移模型应采用脾内注射切脾法。
Objective To establish the colonic carcinoma liver metastases model for gene expression prof iles in BALB/C mice. Methods Liver metastases were established in BALB/C mice by intrasplenic injection of colonic carcinoma cells(C26) . Divided into Spleen removal group and spleen Reservations group.To observe the survival time.The liver metastases conditions were dissected and recorded for all animals. Results Spleen removal group,The incidence rates of lung metastases was 100%,Survival time was 11~15 days,the average survival time of tumor bearing animals was(13.05±0.71) days. spleen Reservations group,The incidence rates of liver metastases was 100%,Survival time was 9~15 days,the average survival time of tumor bearing animals was(11.64±1.49) days. Spleen removal group,liver tumor completely occupied the leaves. spleen Reservations group,a huge tumor in the spleen,liver metastasis mostly scattered in the grain size of the tumor nodule metastasis. Conclusion Gene expression prof iling of colon cancer in mice for liver metastasis model should be used spleen removal injection method.
