RIM-19的表达对人子宫颈癌细胞移植瘤血管生成的影响

摘要 目的:通过裸鼠体内实验探讨GRIM-19对子宫颈癌细胞血管生成的影响。方法:采用western blot检测GRIM-19和VEGF的mRNA和蛋白水平表达情况。将HeLa/Control、HeLa/GRIM-19接种到裸鼠皮下检测荷瘤情况,利用免疫组化法计数微血管密度。结果:HeLa/GRIM-19细胞接种肿瘤生长速度明显减慢(P<0.01)。移植瘤蛋白检测发现GRIM-19表达升高,而VEGF的表达降低。HeLa/Control移植瘤的微血管密度是72±9.3/mm2,而HeLa/GRIM-19的微血管密度为32±4.7/mm2,两者差异有统计学意义(P<0.05)。结论:GRIM-19表达可以抑制子宫颈癌细胞的微血管生成,可能是子宫颈癌治疗的新靶点。 Objective:To explore the effect of GRIM-19 expression on angiogenesis of transplanted tumor of cervical cancer cells in vivo in nude mice.Methods:The expression levels of mRNA and protein of GRIM-19 and VEGF were detected by western blot.HeLa/Control and HeLa/GRIM-19 were inoculated into skin of nude mice to detect tumor-bearing condition,the microvessel density was measured with immunohistochemical method.Results:The growth rate of HeLa/GRIM-19 transplanted tumor slowed down significantly(P0.01).The expression of GRIM-19 protein increased,while the expression of VEGF decreased in protein detection of transplanted tumor.The microvessel density of HeLa/Control transplanted tumor was 72±9.3/mm2,while the microvessel density of HeLa/GRIM-19 was 32±4.7/mm2,there was significant difference between the two groups(P0.05).Conclusion:The expression of GRIM-19 can inhibit the angiogenesis of microvessels of cervical cancer cells,which may be a new target for treatment of cervical cancer.
