摘要
目的研究细胞因子IFN-γ在小鼠不同妊娠阶段中的免疫调节作用。方法用ELISA检测不同孕期小鼠子宫和肝脏组织匀浆中IFN-γ的表达,通过免疫组织学方法观察IFN-γ清除对小鼠uNK细胞数量的影响。结果 ELISA检测后发现子宫分泌的IFN-γ与uNK细胞数的动力学变化特征呈伴随关系,并且其分泌量均低于同期肝脏的分泌水平。由于IFN-γ对uNK细胞的扩增有重要的作用,IFN-γ清除鼠的uNK细胞数量明显少于对照鼠。结论子宫NK细胞有大量表达IFN-γ的潜能,正常情况下仅适量分泌以维持Th1/Th2之间的平衡,保证妊娠的顺利进行。
Objective To compare the differences of cytokines secretion of IFN - γ by NK cells in uterine ( uNK cells) and different organ - derived murine NK cells, and focus on the cytokines secretion by uNK cells. Methods Mechanical dissection approaches were used to prepare cell suspension. The cytokines production of IFN - γ in different tissues during pregnancy were detected by enzyme - linked immu- nosorbent assay (ELISA)? Histological study was also conducted to confirm the uNK population after anti -IFN -γ blocking, by periodic acid - Schiff (PAS) staining? Results It was demonstrated that the expression of IFN -γ in uterine endometrium of pregnant mice were lower than in liver, but increased significantly during pregnancy till mid gestation day? Uterine NK cells had more IFN -γ production ability than other organ- derived NK cells? Anti IFN -γ treatment decreased the local uNKcells number in pregnant mice. Conclusion Uterine NK cells have a potential of producing abundant IFN - γ, but a few cytokines were produced in order to banlance the content of Thl/Th2 type cytokine during the normal pregnancy of mice, thus ensuring successful pregnancy progress.
