摘要
目的研究420nm强脉冲光对痤疮动物模型的疗效观察及其作用的可能机制。方法皮内注射痤疮丙酸杆菌构建大鼠耳廓炎症痤疮模型,用420nm强脉冲光照射痤疮动物模型,观察照射前后炎症皮损的改变,进行皮肤病理学检查,免疫组化法和Real-timePCR法测定TNF-α及MMP-2等因子的表达水平。结果模型组注射部位于第5天形成痤疮样皮损;照射组与模型组相比,炎症反应明显减轻;病理示毛囊角化层明显变薄,真皮层炎性细胞减少,皮脂腺缩小;免疫组化示TNF-α和MMP-2呈阳性表达,且照射使其阳性表达降低;Real-timePCR示TNF-α和MMP-2在模型组中呈高表达,照射后表达下调(P<0.05)。结论 420nm强脉冲光对痤疮有良好的治疗效果,其有效性可能与细胞因子TNF-α及MMP-2等的表达有关。
Objective To study the effect of 420 nm intense pulse light on animal acne model and its possible mechanism. Methods A rat model of auricular acne was established by intradermal injection with propionibac-terium and irradiated with 420 nm intense pulse light. Inflammatory skin damage was observed before and after irradiation. Expression of TNF-α,MMP-2 and other factors was detected by dermatopathology,immunohistochemistry and real-time PCR,respectively. Results Acne-like lesions were observed in acne model group 5 days after injection of propionibac-terium. The inflammatory reaction level was significantly lower,the folliculus pili cuticular layer was notably thinner,the number of dermal inflammatory cells was obviously less,and the sebiferous gland was remarkably smaller in irradiation group than in control group. Immunohistochemistry showed positive expression of TNF-α and MMP-2 in model group,which was influenced by irradiation. Real-time PCR showed over-expression of TNF-α and MMP-2 in model group,which was reduced after irradiation ( P 0. 05). Conclusion 420 nm intense pulse light has good therapeutic effects on ance,which may be related with the expression of TNF-α,MMP-2 and other factors.
