摘要
目的:探讨冠心病患者外周血中肝细胞生长因子浓度与CD133+内皮祖细胞数目的关系及临床意义。方法:30例冠心病患者(冠心病组)与30例非冠心病患者(非冠心病组),采用密度梯度离心法获取外周血单个核细胞,以FITC-鼠抗人CD133标记,流式细胞仪检测其CD133+细胞数目;用ELISA法检测血浆中肝细胞生长因子浓度。结果:与非冠心病组比较,冠心病组外周血中CD133+细胞数目少,血浆中肝细胞生长因子浓度高,差异均有统计学意义(P<0.01)。冠心病患者外周血中CD133+细胞数量与肝细胞生长因子浓度呈负相关。结论:检测外周血中CD133+细胞和血浆中肝细胞生长因子浓度,有望成为诊断冠心病的辅助检查。
Objective To explore the relationship between hepatocyte growth factor and the numbers of CD133+ cells in peripheral blood in patients with coronary heart disease and the clinical significance.Methods Mononuclear cells were obtained from peripheral blood by density gradient centrifugation in 30 patients with coronary heart disease and 30 patients without coronary heart disease,and marked with FITC-conjugated mouse anti-human CD133 monoclonal antibody.Then the number of CD133^+ cells was detected with flow cytometry.The concentration of hepatocyte growth factor protein in blood plasma was determined with ELISA techique.Results Compared with the controls,the number of CD133^+ cells in peripheral blood of patient with coronary heart disease was lower,and the concentration of hepatocyte growth factor protein in blood plasma was higher,which showed significant differences.The number of CD133+ cells was negatively correlated to the concentration of hepatocyte growth factor(P〈0.01).Conclusion It is a promising auxiliary diagnosis for coronary heart disease to detect the number of CD133+cells and the concentration of hepatocyte growth factor in periheral blood.
