摘要
目的对神经毒素1-甲基-4苯基-1,2,3,6-四氢吡啶(MPTP)引起的中脑黑质细胞死亡本质进行研究。方法应用MPTP腹腔注射C_57-BL小鼠,分别用DNA末端转移酶介导的dUTP-x缺口末端标记法(TUNEL)和流式细胞术法(FACS)定性和定量检测黑质细胞凋亡。结果制备C_57-BL小鼠帕金森病(PD)模型的MPTP常规剂量(30mg/kg)连续注射7d明显地导致黑质细胞凋亡,连续注射3d轻度引起黑质细胞凋亡。低剂量MPTP(10mg/kg)连续注射7d未能引起黑质细胞凋亡,此剂量不能制备PD小鼠模型。结论由MPTP制备的PD小鼠模型,导致中脑黑质多巴胺能神经元毁损的本质可能是细胞凋亡。
Objective To study the nature of 1 - methyl - 4 - phenyl - 1,2,3,6 -- tetrahydropyridine (MPTP) - induced mesencephalic nigral neuronal death. Methods C_57BL mice were treated with MPTP, TUNEI and flow cytometry (FACS) were applied to detect the neuronal apoptosis in the substantia nigra. Results The administration of MPTP 30mg/kg for seven days remarkably or for three days mildly induced nigral neuronal apoptosis in mice. These doses were at least required to produce behavioral abnormalities and marked reduction of dopamine in the nigrostrlatal system. However, MPTP 10mg/kg for 7 days could not lead to the nigral apoptosis, this low dose was not able to develop the changes of behavior and neurochemistry. Conclusion MPTP is a substance for developing animal model of PD, and apoptosis may be a main factor for MPTP's injuring dopaminergic neurons in the substantia nigra.
