摘要
目的研究河北省未接受抗HIV治疗患者体内HIV毒株耐药突变情况,为开展临床抗病毒药物治疗和制定合理的用药方案提供数据支持。方法 RT-PCR技术扩增HIV pol区基因,双脱氧法测定HIV基因序列,并使用Contig Express软件处理原始核苷酸序列,与Stanford Hiv Drug Resistance Database(hivdb.stan-ford.edu)进行对比分析耐药突变情况。结果 10例患者HIV毒株基因亚型分析结果均为B亚型。发现存在大量逆转录酶和蛋白酶抑制剂继发耐药变异。结论河北省未治疗患者体内HIV继发耐药变异的存在提示要及早进行耐药检测,适时调整治疗方案。
Objective To study the drug resistance mutations of HIV-1 strains in untreated HIV/AIDS patients in Hebei province, and to provide the base-line data for clinical antiviral treatment and suitable therapeutic schedule. Methods The partial pol sequences were amplified by RT-PCR , then the gene segment was sequenced. The results of HIV genic sequence were treated by Contig Express and were compared with the Stanford Hiv Drug Resistance Database to analyze the state of HIV drug resisitance. Results ( 1 ) Phylogenetic analysis indicated that HIV genic subtypes of 10 patients were classified as subtype B. (2) Lots of secondary mutations were related with RT inhibitor and PI inhibitor. Conclusion The results reveals that untreated patients with HIV have acquired many secondary mutations in their PR and RT genes , which suggests us to pay much attention to the detection of drug resistance mutations and the adjustment of suitable therapeutic schedule in time.
