摘要 目的探讨舌癌细胞对淋巴管内皮细胞(LECs)表型及功能特性的影响以及LECs在舌癌淋巴道转移中的作用。方法 LECs与舌癌细胞进行共培养,模拟肿瘤中LECs的变化。对肿瘤中LECs的生物学行为,如增殖、淋巴管生成及凋亡进行检测。同时,在转录水平对与淋巴管内皮细胞生物学行为改变相关的一组基因应用荧光实时定量PCR进行检测。结果与LECs比较,肿瘤中LECs具有更高的增殖活性、淋巴管生成及抵抗凋亡的能力。肿瘤中LECs中EDIL3、NRP1、ANGPTL4、VEGFR1、VEGFC、VEGFA及FN1在转录水平基因表达明显上调。结论在舌癌细胞的影响下,淋巴管内皮细胞的生物学行为发生了改变,并且伴随着基因表达差异,可能会促进舌癌细胞的淋巴道转移。 Objective To investigate the effects of tongue cancer cells on the phenotype and functional properties of lymphatic endothelial cells(LECs)and explore the LECs function in lymphatic metastasis of tongue cancer cells.Methods LECs were co-cultured with tongue cancer cells to analogue the changes of LECs in tongue cancer tissue.The biologic behaviors of tumor-derived LECs(TLECs)including proliferation,lymphangiogenesis and apoptosis,were compared with those of LECs.The differential expression of genes was confirmed by real-time PCR.Results TLECs were more proliferative,had more resistance to apoptosis and enhanced ability to organize capillary-like structures compared to LECs.The mRNA expression of EDIL3,NRP1,ANGPTL4,VEGFR1,VEGFC,VEGFA,and FN1 were upregulated in TLECs.Conclusion Under the influence of tongue cancer cells,the biologic behaviors of LECs altered and LECs had different gene expression pattern,which might contribute to the lymphatic metastasis of tongue cancer cells.
