摘要
通过盐溶液溶胀和酶液消化制备小肠黏膜下层基质作为血管组织工程支架材料,将大鼠内皮细胞接种于支架材料上,测定小肠黏膜下层基质的细胞相容性.实验结果表明:内皮细胞在小肠黏膜下层基质上迅速铺展增殖生长,在4 h时大约有96%的细胞黏附于材料上,MTT法结果表明内皮细胞在支架上增殖生长明显,活细胞形态学观察表明细胞在支架材料上生长良好.
As the scaffolds of the vascular tissue engineering,the small intestinal submucosas matrix are obtained by salt solution swelling and enzyme digestion,and vascular endothelial cells are isolated from the rat thoracic aorta and subcultureed and purified.The ECs are seeded on the surface of scaffolds to determine whether the scaffolds had the cytocompatibility.Experimental results show that the seeded endothelial cells proliferate rapidly on the small intestinal submucosa matrix.About 96% of the cells adhere to the surface of the material in the 4 hours after being seeded,and the Ecs grow obviously on scaffolds by the MTT method.The morphological observation of cells shows that cells grow well on the scaffold materials.
