摘要
目的:探讨解聚复肾宁(黄芪、地黄、黄连、丹参、姜黄、葛根等)(JJFSN)治疗早期糖尿病肾病(DN)的作用机制。方法:高糖诱导系膜细胞(MC)增殖以及细胞外基质(ECM)分泌,制备不同浓度JJFSN含药血清作用于体外培养的MC,用MTT法检测细胞增殖,放射免疫技术(RIA)检测细胞培养上清中Ⅳ型胶原(ColⅣ)及层黏连蛋白(Ln)的含量。结果:JJFSN含药血清可抑制高糖诱导的MC过度增殖(P<0.05),并且这种作用具有药物剂量和时间依赖关系。同时,JJFSN可逆转高糖对ColⅣ和Ln分泌的影响,降低MC培养上清中ColⅣ和Ln的含量(P<0.05),并且这种作用随着JJFSN剂量的增加而增强。结论:JJFSN抑制高糖诱导的MC过度增殖并减少ECM的过量分泌,可能是其防治DN早期的作用机制。
AIM: To investigate the effect of a traditional Chinese compound,Jiejufushening (Radix astragli;Radix rehmanniae;Rhizoma coptidis;Radix et Rhizoma salviae multiorrhizae;Rhizoma Curcumae longae;Radix puerariae lobatae,etc) (JJFSN) on proliferation and secretion of extracellular matrix(ECM) of rat mesangial cells (MC).METHODS: Sera containing different dose of JJFSN were prepared and put together with 25mM glucose.MC was cultured in vitro for 12 h,24 h and 48 h respectively.The effects of the drug-containing sera on MC cell proliteration were assessed by the methylthiazoletetrazolium(MTT) assay.The supernatants were collected after 48 h growth.And collagen type Ⅳ ( Col Ⅳ) and laminin ( Ln) were detected by radioimmunoassay ( RIA).RESULTS: JJFSN pharmacological serum significantly(P 0.05) inhibited MC proliferation under high glucose condition,in a dose and time-dependent manner.Meanwhile,compared with the high glucose group,the concentration of Col Ⅳ and Ln declined in different concentration JJFSN groups(P 0.05),in a dose-dependent manner.CONCLUSION: JJFSN can supress the influence of high glucose on MC proliferation and secretion of Col Ⅳ and Ln.It implies that JJFSN might be a potential therapeutic agent for the prevention against DN in the early stage.
