摘要
考察了10种水溶性有机溶剂对HPLC测定头孢菌素C含量的影响,采用C_(18)色谱柱,流动相为磷酸二氢钠-甲醇(90:10),波长254 nm。发现不同的有机溶剂有不同程度的影响,其中乙醇的影响最大;乙醇浓度越高,峰面积就越小。将含有乙醇的头孢菌素C溶液稀释到一定程度,能够有效消除乙醇的影响。稀释法具有较好的精密度、重复性和回收率,为在工业生产中准确测定头孢菌素C的含量提供了依据。
The influences of ten kinds of organic solvents on the determination of cephalosporin C by HPLC were investigated. Using C18 reverse phase column, mobile phase was phosphate buffer-methanol (90:10) and the detection wavelength was 254 nm. The results showed that different organic solvents influenced the determination of eephalosporin C by HPLC to different extents, among them ethanol had the greatest impact. The higher of the ethanol concentration, the less peak area of cephalosporin C was. It could effectively eliminate the influence of ethanol by diluting the cephalosporin C solution to certain extent . The dilution method had good precision, reproduction and recovery, which provided a basis for the accurate determination of cephalosporin C in industrial production.
