儿童钙代谢相关激素与Ⅰ型胶原及骨钙素基因多态性关系的分析

摘要 目的探讨Ⅰ型胶原及骨钙素基因多态性与儿童钙代谢相关激素之间的关系。方法以河南省某地8~12岁中国汉族健康儿童作为研究对象,采用放免法测定血清骨钙素(OC)和降钙素(CT);采用PCR-RFLP技术检测COL1A2 PvuⅡ、RsaⅠ和OC HindⅢ基因多态性。结果携带RR基因型的儿童血清Ca和CT浓度均值分别为2.75mmol/L和206.42ng/ml,均高于rr基因型儿童的2.36mmol/L和170.15ng/ml(分别P<0.05)。血清OC浓度在COL1A2 RsaⅠ各基因型未见差异(P>0.05),也未发现COL1A2 PvuⅡ及OC HindⅢ基因型与儿童血清Ca、CT及OC浓度相关。结论 COL1A2 RsaⅠ多态性可能对儿童血清钙及降钙素水平产生影响。 Objectives To investigate the relationship between polymorphisms in the collagen type I alpha 2 (COL1A2),osteocalcin (OC) gene and serum calciotropic hormone levels.Method One hundred twenty-two healthy children aged 8-12 years old were recruited.The Pvu Ⅱ and Rsa Ⅰ polymorphisms in the COL1A2 gene and Hind Ⅲ polymorphism in the OC gene were determined using the PCR-RFLP procedure.Calcitonin(CT) and OC levels in the serum were measured using radioimmunassays.Results The calcium and CT in serum of children carrying the homozygous genotype RR were found to be significantly higher compared to children carrying the homozygous genotype rr (P0.05).However,the differences in calcium,CT and OC levels were not observed in the Pvu Ⅱ polymorphism in COL1A2 gene and Hind Ⅲ polymorphism in the OC gene.Conclusion This study provides the evidence of an association between polymorphisms in the COL1A2 gene with calcium and calcitonin in serum of children.
