摘要
目的探讨Snail和E-cadherin表达与肝细胞癌转移的关系及干扰Snail对肝癌细胞转移的影响。方法采用免疫组化和RT-PCR分别检测36例肝癌组织中Snail,E-cadherin蛋白和mRNA的表达。检测HCC9024细胞中Snail对E-cadherin mRNA和蛋白表达的逆转作用及对HCC9024体外侵袭、运动的抑制作用。结果转移组肝癌组织中Snail蛋白和mRNA表达明显高于非转移组(P〈0.05),而E-cadherin在转移组肝细胞癌中无表达。Snail mRNA在未转染组、瞬时转染组、稳定转染组中的表达为0.985±0.016,0.554±0.011,0.206±0.017,P〈0.05。E-cadherin mRNA在未转染组、瞬时转染组、稳定转染组中的表达为0.120±0.001,0.360±0.002,0.727±0.006,P〈0.05。Snail和E-cadherin蛋白表达结果与mRNA表达结果一致。HCC9024细胞体外运动和侵袭能力实验表明,未转染组、阴性对照组及转染Snail组跨膜细胞数及穿透基底膜细胞数差异有统计学意义(P〈0.05)。结论肝癌细胞中Snail mRNA可影响E-cadherin mRNA的表达,并在肿瘤细胞获得侵袭和转移能力的过程中起重要作用,阻断Snail mRNA的表达可能会为肝癌的基因治疗提供新的靶点。
Objective To investigate the expression of Snail and E-cadherin in hepatocarcinoma tissues and the inhibitory effects of anti-Snail on invasion and migration of hepatocarcinoma cells. Methods Snail and E- cadherin protein and mRNA were analysed by ICH and RT-PCR in 36 cases of hepatocarcinoma patients. To de- tection the Snail's reverse effect on E-cadherin mRNA expression and the invasion and motility in vitro in HCC9024. Results The expression of Snail protein and mRNA in metastasis hepatocarcinoma tissue is higher than that of in non-metastasis, and E-cadherin mRNA is lower in metastasis tissues( P 〈 0.05). Snail mRNA in non-transfeeted group, transient transfection group, stable transfection group were 0. 985 ±0. 016,0. 554±0.011, 0. 206 + 0.017 (P 〈 0.05 ) ; E-cadherin mRNA in non-transfected group, transient transfection group, stable transfection group were 0. 120 ± 0. 001,0. 360 ±0. 002,0. 727 ±0. 006 ( P 〈 0.05 ). Snail and E-cadherin protein expression results were consistent with the mRNA. HCC9024 cell movement and invasion in vitro experi- ments showed that non-transfected group, negative control group and Snail transfected group' s transmembrane and penetrate the basement membrane cells also had statistically significant differences ( P 〈 0.05 ). Conclu-sions Snail mRNA in the hepatocarcinoma cells may affect E-cadherin mRNA expression, and play an important role in tumor cell invasion and metastasis, It may provide new targets for gene therapy of hepatocarcinoma.
