摘要
目的:调查HPA-1抗原系统a/b/x等位基因在汉族人群中的多态性分布。方法:利用聚合酶链反应-序列特异引物技术(PCR-SSP)技术对1000例中国无关健康汉族人群样本进行HPA-1抗原系统等位基因分型,随机抽取部分样本进行测序验证。结果:本次调查共检测到HPA-1a/a基因型988个,HPA-1a/b基因型12个,未检测到HPA-1x等位基因。结论:HPA-1抗原系统在中国汉族人群中以HPA-1a等位基因为主(99.4%),HPA-1b分布频率较低(0.6%)。目前国内尚未发现HPA-1x等位基因,有必要进一步扩大样本对不同地区、不同民族进行多态性调查。
Objective:To investigate the polymorphism of HPA-1 antigen system a/b/x alleles in Chinese Han population.Method:HPA-1 antigen system alleles of 1000 samples from unrelated healthy Chinese Han population were genotyped by PCR-SSP(Polymerase Chain Reaction-Sequence Specific Primed),part of samples were selected randomly and sequenced for confirmation.Result:988 genotypes of a/a and 12 of a/b alleles in HPA-1 antigen system were identified,none of HPA-1x allele were found.Conclusion:Of the identified HPA-1 antigen system polymorphisms,HPA-1a allele is primary in Chinese Han population(the frequency is 99.4%),the frequency of HPA-1b polymorphism distribution is low(0.6%).In the present study,none of HPA-1x alleles have identified in China,further investigation on polymorphism distribution in various areas and populations is needed with larger samples.
