摘要
对柿树离体繁殖中外植体的褐变进行了研究,结果表明,外植体褐变与多酚氧化酶活性有关。多酚氧化酶活性与柿树的物候期有密切联系,2月份取即将萌动的休眠芽进行培养,褐变死亡率低,4月份取嫩枝茎尖进行培养,褐变死亡率高。选择适宜的培养基类型(改良MS培养基),配合适当的激素种类及浓度(ZT913μmol·L-1,IAA057μmol·L-1),能促进外植体的生长与分化,减轻外植体的褐变程度。培养基中加入PVP可有效地防止外植体褐变,提高外植体的分化率和增殖倍数。
In this experiment,the explant browning in the proceeding of vitro propagation of Diospyros kaki Linn. was studied.The results showed that the browning death rate of dormancy buds collecting in Feb.was lower than that of new shoots collecting in Apr.in the culture.The reason for this had some close relationship with the activity of polyphenol oxidase(PPO).Selecting suitable medium(modified MS Medium),and using proper hormone and its concentrations(ZT 9.13 μmol·L -1 .IAA 0.57 μmol·L -1 ),could not only promote the growth and differentiation,but also reduce the degree of browning of explant Adding PVP in the medium could prevent browning effectively and increase the differentiation rate and proliferation times of explant.
