摘要 目的:观察不同浓度葡萄糖对体外培养的MC3T3-E1成骨细胞凋亡的影响。方法:不同浓度的葡萄糖、甘露醇分别刺激体外培养的MC3T3-E1成骨细胞1、3、7、12d,用流式细胞术检测成骨细胞的凋亡。结果:甘露醇组MC3T3-E1成骨细胞凋亡较正常对照组有一定的增加,但无统计学差异(P>0.05);而葡萄糖呈浓度、时间依赖性诱导MC3T3-E1成骨细胞凋亡(P<0.05,P<0.01)。结论:高糖状态下,MC3T3-E1成骨细胞凋亡显著增加。 Objective:To study the influence of different glucose concentrations on the apoptosis of MC3T3-E1 osteoblast. Methods: Seven culture media of different glucose and mannitol concontrations,5.5,11.1,22.2,33.3 mmol/L were set up. MC3T3-E1 osteolblast was cultured in these media successively for 1,3,7,12 d,and then the percentages of apoptotic cells were measured by flow cytometry. Results:High mannitol can slightly increase the apoptosis of MC3T3-E1 osteoblast,but there was no statistical significance(P〉0.05). The inducing action of glucose to the apoptsis of MC3T3-E1 osteoblast was in concentration and time dependent manner(P〈0.05,P〈0.01). Conclusion:Glucose of elevated concentration significantly promotes the apoptosis of MC3T3-E1 osteoblast.
