摘要
目的探讨汉族基质金属蛋白酶-3(matrix metalloproteinase 3,MMP-3)-1171 5A/6A基因多态性与原发性高血压(essential hypertension,EH)伴颈动脉粥样硬化(carotid atherosclerosis,CAS)的关系。方法采用病例-对照研究,应用聚合酶链反应-限制性片断长度多态性(polymerase chain reaction?restriction fragmentlength polymorphism,PCR-RFLP)技术测定MMP-3-1171 5A/6A基因多态性,共检测了新疆医科大学第一附属医院高血压科住院的131例汉族EH伴CAS患者(essential hypertension with carotid atherosclerosis,AS组)和86例汉族EH伴颈动脉内膜正常患者(essential hypertension without carotid atherosclerosis,NS组)。结果 AS组的6A等位基因频率高于NS组(P〈0.05),携带6A等位基因增加了EH伴CAS的危险度(OR=1.885,95%CI:1.013~3.505)。结论 MMP-3-1171 5A/6A基因多态性可能与汉族EH患者伴CAS有关,6A等位基因可能增加了EH患者伴发CAS的风险。
Objective To investigate the relationship between the single nucleotide polymorphisms of matrix metalloproteinase-3(MMP-3)-1171 5A/6A and essential hypertension with carotid atherosclerosis in Chinese Han population.Methods The study comprised 217 Han nationality participants,who were divided into two groups: essential hypertension with carotid atherosclerosis(AS group,n=131) and essential hypertension without carotid atherosclerosis(NS group,n=86).Genotypes were detected by polymerase chain reactions followed by restriction analyses with specific endonucleases and their frequencies were determined.Results Individuals with 6A/6A genotype had 1.885 fold risk suffering from essential hypertension with carotid atherosclerosis,when compared with 5A/5A and 5A/6A genotypes.Conclusion The single nucleotide polymorphism of matrix metalloproteinase-3(MMP-3)-1171 5A/6A is probably associated with carotid atherosclerosis in essential hypertension people of Chinese Han population.
