摘要
用紫茎泽兰70%的乙醇提取物及其氯仿萃取物,对番茄青枯菌生理、生化、细胞形态等方面进行抑菌试验。乙酸乙酯、正丁醇、乙醚、氯仿萃取物具有较好的热稳定性,在pH9时抑菌圈最大;通过透射电镜观察,菌体经紫茎泽兰提取物作用后,鞭毛消失,中央出现空腔。SDS-PAGE显示紫茎泽兰氯仿萃取物处理番茄青枯菌后,菌体内蛋白质减少,随着作用时间的延长,完全抑制或破坏了小分子蛋白质的合成,对大分子蛋白质也有一定的抑制作用。POD、CAT、SDH酶活测定显示,随着加入的紫茎泽兰氯仿萃取液浓度的增大,酶活显著降低。以上试验结果表明,紫茎泽兰氯仿萃取液对番茄青枯菌有明显的抑制作用,对于番茄青枯病防治的应用有一定的指导意义。
70% of the ethanol extract of Eupatorium adenophorum Spreng and its chloroform extract was tested on Ralstonia solanacearum in the aspects such as physi01ogy, biochemistry, cell morphology and metabolism. The experiment showed that ethyl acetate, butanol, ether, and chloroform extract exhibited satisfactory thermal stability, and the largest zone of inhibition was observed when the condition of prig, with electron microscopy, also, the bacterial flagellum disappeared and the central of bacteria turned cavity. SDSPAGE showed after the effects of chloroform extract of Eupatorium adenophorum Spreng,protein of bacteria reduced . As the role of the time extended,micromolecular proteins were completely inhibited or its synthesis were undermined. The experiment also showed that chloroform extract had inhibitory effect against some micromolecular proteins. With the concentration of chloroform extract of Eupatorium adenophorum Spreng was increasing, the enzyme activity of POD, CAT and SDH decreased significantly. These research showed that chloroform extract exhibited best inhibitory effect against Rastonia solanacearum and could provide some guidenee for the application of controlling Rastonia solanacearum.
