摘要
目的探讨化浊解毒调肝方抗肝纤维化的作用机制。方法将120只健康雄性SD大鼠随机分为6组。即正常组、模型组、秋水仙碱预防组、化浊解毒调肝方预防组、秋水仙碱治疗组、化浊解毒调肝方治疗组各20只,除正常组外其它各组均采用腹腔注射未灭活的猪血清建立肝纤维化大鼠模型,分别采用秋水仙碱和化浊解毒调肝方进行药物干预。各组分别于造模8、12周末随机处死10只大鼠,用免疫组化法观察药物对肝组织基质金属蛋白酶抑制因子(TIMPs)表达的影响。结果化浊解毒调肝方有明显的抑制肝纤维化模型大鼠肝组织TIMP-1的表达的作用,且抑制作用要优于秋水仙碱(P<0.05)。结论化浊解毒调肝方可有效防治大鼠肝纤维化,其作用机制可能是通过抑制肝纤维化组织TIMP-1的表达实现的。
Objective To research the mechanism of Huazhuo Jiedu Tiaogan prescription on anti-hepatic fibrosis.Methods 120 health male SD rats were randomly divided into six groups：control group,model group,colchicines prevention group,Huazhuo Jiedu Tiaogan prescription prevention group,colchicines treatment group and Huazhuo Jiedu Tiaogan prescription treatment group,20 rats in each group.The model rats with hepatic fibrosis were induced intraperitoneal injection of un-inactivated porcine serum.Colchicines and Huazhuo Jiedu Tiaogan prescription were adopted to treat.The expression of tissue inhibitor of metalloproteinases（TIMPs）was measured with immunohistochemistry at the No.8 week and No.12 week of establishing model,executing 10 rats.Results Huazhuo Jiedu Tiaogan prescription inhibited obviously the expression of TIMP-1 in hepatic tissue of rat with hepatic fibrosis in comparison with that in colchicines.Conclusion Huazhuo Jiedu Tiaogan prescription can prevent and treat hepatic fibrosis rat,its mechanism maybe inhibit the expression of TIMP-1 in hepatic tissue.
