摘要
以相关文献中查阅的10种液体种子培养基配方作为初始研究培养基,通过比较相应的种子液中菌球的生长状况以及固体发酵产菌丝的情况,筛选出良好的液体种子初始培养基为玉米面1.0%、蔗糖2.0%、蛋白胨0.2%、KH2PO4 0.1%、MgSO4.7H2O 0.05%。以筛选出的液体种子初始培养基进行3因素3水平正交设计,获得了最优的液体种子培养基配方为:玉米面1.5%、蔗糖2.0%、蛋白胨0.1%、KH2PO4 0.1%、MgSO4.7H2O 0.05%、V B 1微量。最终可达到每瓶中的菌球数为6 000,且菌丝在4 d的时间就可基本上生长良好。
Selecting the ten liquid seed culture conditions referred to the related literatures as the primary conditions,it is observed the mycelial pellets in the seed culture medium and the growth of Ganoderma Sinensis hypha by the solid fermentation,then the basic liquid seed medium of corn powder 1.0%,sucrose 2.0%,peptone 0.2%,KH2PO4 0.1%,MgSO4.7H2O 0.05% are confirmed.Also,it is optimized the seed culture medium in a three factor-three level orthogonal experiment,that is,corn power 1.5%,sucrose 2.0%,peptone 0.1%,KH2PO4 0.1%,MgSO4.7H2O 0.05%,and minimumV B 1.As a result,the mycelial pellets in the shaking flask reached 6 000,and Ganoderma Sinensis hypha could grow well in four days in the optimized seed medium.
