雷至胶囊对阿霉素肾病大鼠肾小球系膜基质增生抑制作用的实验研究 被引量:5

摘要 目的:研究雷至胶囊对阿霉素肾病大鼠肾小球系膜基质增生的抑制作用,探讨其抗系膜基质增生的分子机制。方法:采用阿霉素静脉注射建立阿霉素肾病模型,将实验动物分为模型组,雷至胶囊组及正常组,分别留取各组第0、2、3、4、5周尿标本。第5周末处死大鼠,留取血肝肾标本;光镜、电镜观察肝肾组织病理形态学变化;West-Blot检测肾组织TGF-β、MMP-9蛋白表达。结果:与模型组相比,雷至胶囊组24 h尿蛋白、血脂及系膜基质指数明显降低,同时TGF-β蛋白表达明显降低,MMP-9蛋白表达明显升高,透射电镜显示肾小球基质增生明显改善。结论:雷至胶囊可从多方面、多层次恢复细胞外基质(ECM)代谢平衡,防止系膜基质增生。 Objective:To investigate the effect of Leizhi capsule on renal mesangial matrix in adriamycin(ADR)-induced nephropathy rats and to explore the mechanism of this effect.Methods:Eighteen SD rats were randomly divided into the nephropathy model group,Leizhi capsule group and normal control group.The rats in the first two groups were induced by intravenous injection of adriamycin on day 1 and day 7.The urine samples were collected at week 0,2,3,4,5 after first adriamycin injection respectively.At the end of the 5th week,the rats were sacrificed and the renal tissue was acquired for the examinations of light microscopy,electron microscopy and protein expression of TGF-β and MMP-9.Results:Compared with those in the rats in the model group,24 h urinary protein,blood cholesterol/triglyceride,mesangial matrix index and the protein expression of TGF-β in the rats of Leizhi capsule group were decreased,albumin and the protein expression of the protein expression of MMP-9 were increased,the examination of transmission electron microscopy showed that the injury on foot processes of podocyte was relieved in rat by administration of Leizhi capsule.Conclusion:Leizhi capsule can restore ECM metabolic balance in multiaspects,multi-level,preventing the proliferation of renal mesangial matrix.
