摘要
以家蚕丝素为载体,采用交联-吸附法固定化单宁酶。正交试验结果表明,最佳的固定化条件为戊二醛浓度为0.45%,固定化pH值为5.0,给酶量15mg/g载体,固定化时间9 h,固定化温度10℃。与游离酶相比,该固定化单宁酶的热稳定性明显提高,25℃贮存6d后酶活仍保持57.5%。以没食子酸甲酯为底物,固定化单宁酶经连续6次酶促反应后,酶活仍保持77.4%。
Tannase was immobilized on silk fibroin (SF) using glutaraldehyde (GA) as cross-linking reagent. The orthogonal tests indicated that the optimum immobilization conditions were as follows: 15 mg tannase were added to 1.0 g activated SF by 0.45% GA, and immobilized at pH5.0 and 10℃ for 9h. Compared with free tannase, the thermal stability of immobilized tannase was obviously improved. The activities of immobilized tannase were still kept to 57.5% when stored at 25℃ for 6 d, and was retained 77.4% after consecutive reaction of 6 times.
