摘要 目的探讨SiRNA-EGFR对人乳腺癌细胞乳腺癌表皮生长因子受体表达的影响。方法应用RT-PCR测定RNA干扰人乳腺癌细胞MDA-MB-231、MDA-MB-453S、ZR-75和ZR-75-30后Hyase基因的mRNA的表达变化。结果 SiRNA-EGFR作用于表达不同水平透明质酸酶的人乳腺癌细胞株MDA-MB-231、MDA-MB-453S、ZR-75-30和ZR-75后能明显抑制其乳腺癌表皮生长因子受体的mRNA表达水平(P<0.05)。结论 SiRNA-EGFR作用于人乳腺癌细胞后能特异性抑制EGFR的mRNA的表达水平。 Objective To study the effect of SiRNA-EGFR on the expression of hyaluronidase gene in human breast cancer cells.Methods Reverse transcription-polymerse chain reaction was used to detect the changes in the expression of EGFR mRNA in human breast cancer cell lines MDA-MB-231,MDA-MB-435S,ZR-75 and ZR-75-30 after transfection by SiRNA-EGFR.Results After transfection with SiRNA-EGFR,the expression levels of EGFR were significantly inhibited in MDA-MB-231,MDA-MB-435S,ZR-75 and ZR-75-30 cells(P0.05).Conclusion Transfection by SiRNA-EGFR can inhibit the expression of EGFR mRNA in human breast cancer cells.
