摘要
为探讨IVM液中FSH+LH添加方式、半胱氨酸和表皮生长因子(EGF)不同浓度对猪IVM卵母细胞早期孤雌发育的影响,将在不同成熟体系中IVM44 h后的卵母细胞经化学激活后放入NCSU-23中培养,分别于第2、7天统计分裂率和囊胚发育率。结果表明:①IVM液添加有FSH+LH且在培养22 h后更换添加有FSH+LH的IVM液继续培养22 h,其分裂率(83.76±9.95)%和囊胚发育率(23.49±7.74)%显著高于添加FSH+LH连续培养44 h、中间不换液的处理(P<0.05);②IVM液中添加1.2 mmol/L半胱氨酸的囊胚发育率(28.13±10.9)%显著高于添加0、0.3、0.6 mmol/L半胱氨酸的处理,差异显著(P<0.05);③IVM液中添加10 ng/mL EGF的囊胚发育率(23.49±7.74)%显著高于其他浓度处理(P<0.05)。可见:在该研究条件下,IVM液添加有FSH+LH且在培养22 h后更换添加有FSH+LH的IVM液继续培养22 h,并添加1.2 mmol/L半胱氨酸、10 ng/mL EGF,有利于促进猪IVM卵母细胞早期孤雌发育。
The objectives of this study were to investigate the effect of supplementation method of gonadotrophin and different concentrations of cysteine and EGF on prophase parthenogenesis of porcine oocytes in vitro maturation.After in vitro maturation for 44 hours and chemically parthenogenetic activation,the porcine oocytes were cultured in NCUS-23 medium for 7 days,and the cleavage rate was examined on 2th day and the blastula rate was examined on 7th day.The results showed that the cleavage rate and the blastula formation rate of oocytes as changing new maturation medium with FSH +LH at 22 h during the 44-48 h in vitro maturation period,were significantly higher(P0.05) than those of the group of non changing new medium.The blastula formation rate of oocytes for adding 1.2 mmol/L cysteine group(28.13 %) was significantly higher than those of adding 0,0.3 and 0.6 mmol/L cysteine groups(19.31 %,15.46 % and 21.27 %,respectively)(P0.05).The blastula rate of oocytes for group of adding 10 ng/mL EGF was significantly higher than that of the other groups(P0.05).In conclusion,the way of adding FSH+LH and changing new maturation with FSH+LH at 22 h in vitro maturation,and adding 1.2 mmol/L cysteine and 10 ng/mL EGF,could improve the prophase parthenogenesis of porcine oocytes in vitro maturation.
