摘要
目的:探讨NDRG2蛋白在非小细胞肺癌(NSCLC)组织中的表达及其在病理分类、分级、TNM分期中的差异性,为进一步研究NDRG2在NSCLC中的作用及其机制提供依据。方法:采用免疫组化EnVinsion法检测NDRG2蛋白在155例非小细胞肺癌中的表达,统计学分析其病理分类、分级及临床TNM分期的差异性。结果:NDRG2阳性表达主要定位于胞质中和胞膜上,在癌和相应正常肺组织中总阳性率分别为63.23%、100%,两者相比差异性非常显著(P=0.0000),其中肺鳞癌和腺癌阳性率分别为54.65%、73.91%,与正常组织比差异性均非常显著(P=0.0000);鳞癌与腺癌间的阳性率和阳性强度差异性显著(P=0.0131),鳞癌高、中、低分化阳性率分别为87.10%、41.3%和11.11%,经Kruskal-Wallis H检验差异性显著(P=0.0000),腺癌的高、中、低分化的阳性率分别是100.0%、88.89%和31.58%,经检验差异性非常显著(P=0.0000);鳞癌、腺癌的TNM分期Ⅰ、Ⅱa、Ⅱb、Ⅲa、Ⅲb的阳性率有逐渐降低的趋势,经检验其差异性显著,P值分别为0.0005、0.0342。结论:NDRG2在NSCLC中低于相应正常组织中的表达,表达的高低与病理分级高低成正比、与TNM分期高低成反比,很可能对NSCLC发生、发展产生重要影响。
Objective: To investigate the expression of NDRG2 protein and analyze the relationship between NDRG2 expression and pathological types, grades and TNM stages in non - small cell lung cancer (NSCLC). Methods:Tumor samples from 155 cases with NSCLC were collected to detect the expression of NDRG2 protein using antibodies against human NDRG2 protein by immunohistochemical staining(EnVinsion). Differences of protein expression among pathological types, grades and TNM stages in cancers were analyzed statistically. Results: NDRG2 positive expression was mainly located in cytoplasm. NDRG2 positive stainings in NSCLC and corresponding normal tissues were 63.23% ,100.00% (P =0.0000). The samples with NDRG2 positive staining were observed to cover 54.65% and 73.91% in squamous cell lung cancers and adenocarcinomas respectively(P =0.0131 ). The positive NDRG2 staining samples were 87.10% ,41.30% ,72.22% in high differentiated,moderately differentiated and low differentiated squamous cell lung cancers respectively(P =0.0000) ,while positive staining covered 100.0% ,88.89% ,31.58% in highly,moderately and low differentiated lung adenocarcinoma respectively ( P = 0. 00130). Significant differences of NDRG2 positive staining among different TNM stages in squamous cell lung cancers and lung adenocarcinomas re- spectively were also observed by Kruskal - Wallis H test ( P = 0.0005,0. 0342 ). Conclusion: NDRG2 protein levels are decreased in NSCLC compared with corresponding normal lung tissue, and the expression is correlated with pathological types, grades and TNM stages, namely, NDRG2 protein expression levels become direct ratio to cancer differentiated degree and inverse ratio to TNM stages in NSCLC ,which provides clinic evidence for the effects of NDRG2 on cancers.
