摘要
通过荧光光谱、热变性、黏度测定、盐效应实验研究了镧(Ⅲ)-氧氟沙星配合物与小牛胸腺DNA(CT-DNA)的结合作用.结果表明CT-DNA能使配合物的荧光产生猝灭,其猝灭常数K15℃=1.62×10-5L.mol-1,K25℃=1.12×10-5L.mol-1,结合位点数分别为0.9887和1.0324,猝灭机理为静态猝灭.同时,该配合物可使DNA的黏度降低,热变性温度升高,说明该配合物主要以部分嵌插的作用方式与DNA结合.盐效应表明该配合物与DNA之间还存在静电作用.
Binding of La(Ⅲ)-ofloxacia complex with calf thymus DNA was investigated by fluorescent spectroscopy,melting temperature,DNA viscosity titration,and salt effect test.The results show that the fluorescence intensities of the complex decrease in the presence of DNA and the static quenching constant K15℃and K25℃ turns to be 1.62×10^-5 L·mol^-1 and 1.12×10^-5 L·mol^-1.DNA shows an increasing melting point and a decreasing viscosity with the addition of the complex,and this suggests that the complex may interact mainly with calf thymus DNA by partial intercalation.It also indicates that the complex is bound to DNA by electrostatic interaction according to salt effect.
